Yes, but host.def is optionnal. If not present, imake will pick up 
default values from other configuration files (xorg.cf, X11.tmpl, 
Imake.tmpl).
xorgsite.def shows most of the variables that can be set in host.def.

PS. in XFree86 xorg.cf is called xfree86.cf and xorgsite.def is 
xf86site.def.
